%X Through the method of being frozen in the liquid nitrogen for a short time, and then thawing in the room temperature for several times, the protoplasts of <em>Chlamydomonas reinhardtii</em> were broken and chromatin were obtained.Then we exactly transformed the chromatin to the explants of the tobacco leaves by the micromanipulation, and then we cultured the explants, observed by the fluorescent microscope every day.The result indicated that the chromatin transformed explants, the nuclei of <em>C. reinhardtii</em> and tobacco were all changed.At the same time, the explants formed the callus and then grew into gemmule.Through the RT-PCR detection, we found the expression of the nuclear gene rbcs2.
